At the pituitary, the peptide binds the growth hormone-releasing hormone receptor on somatotroph cells. Receptor activation raises intracellular cyclic AMP and triggers release of stored growth hormone. Somatostatin and other hypothalamic signals modulate this response. Negative feedback from insulin-like growth factor 1 also influences output. The same regulatory architecture operates with the native hormone. Whether the synthetic analog alters feedback dynamics over repeated exposure remains an open question. Most published receptor work uses cell models rather than intact human systems.
CJC-1295 is a synthetic peptide belonging to the growth hormone-releasing hormone analog family. It comprises twenty-nine amino acid residues derived from the N-terminal region of natural GHRH. The molecule incorporates several non-natural substitutions that increase resistance to enzymatic degradation. These modifications extend its activity compared with the native hormone fragment. Researchers use it to study pituitary growth hormone secretion in laboratory and clinical settings. This compound is distinct from native GHRH in its stability profile.
Two related forms circulate in technical discussion under the same family name. The original version carries a drug affinity complex (DAC) that binds covalently to serum albumin after administration, and this linkage substantially extends circulation time. A second form, frequently written as modified GRF(1-29) or CJC-1295 without DAC, lacks that linker and clears much faster. The naming is a frequent source of confusion because the shorthand CJC-1295 can refer to either form depending on the source. Reports sometimes fail to specify which variant was studied.
The peptide backbone includes a D-alanine at position two, which resists cleavage by dipeptidyl peptidase IV, and several other substitutions that reduce degradation. Its molecular weight is roughly 3.4 kDa without the linker and about 3.6 kDa with it. The molecule is water soluble and is normally supplied as a lyophilized powder. Precise sequence and mass values depend on which variant is described, so technical documents usually state the exact form being referenced.

Reports on this compound commonly follow serum growth hormone and insulin-like growth factor 1 across defined time windows. Protocols differ in sampling frequency, assay platform, and participant characteristics, which makes direct comparison between publications difficult. Some work focuses on pulsatile release patterns instead of average concentrations. Whether repeated exposure alters endogenous hormone rhythms over long periods remains an open question, and the formal literature is thinner than the volume of informal commentary implies.
Once in circulation, the peptide binds the growth hormone-releasing hormone receptor displayed on pituitary somatotroph cells. Receptor activation couples to Gs proteins, elevates intracellular cyclic AMP, and drives protein kinase A signaling inside the cell. That cascade increases discharge of growth hormone into the bloodstream. The analog therefore operates through a receptor pathway that already exists for the body's own releasing hormone, rather than through an engineered artificial target.
Downstream of growth hormone, the liver and other tissues increase production of insulin-like growth factor 1, a mediator of many growth-promoting effects. Studies have documented elevated levels of both hormones after dosing, and the rise from the long-acting form persists longer than that produced by shorter-acting analogues. What remains unclear is whether sustained elevation of these markers translates into meaningful clinical benefit, and whether prolonged exposure carries risks that short trials could not detect.
CJC-1295 acts at the growth hormone-releasing hormone receptor, a G-protein-coupled receptor found on somatotroph cells in the anterior pituitary. Binding triggers a rise in cyclic AMP and calcium entry, which promotes release of stored growth hormone. Because the peptide mimics the body's own releasing hormone, it amplifies existing secretory pulses rather than driving continuous output. The size of the response therefore depends partly on the subject's own hormonal rhythm and feedback state.
Identity and purity are assessed mainly by reversed-phase high-performance liquid chromatography combined with mass spectrometry. The chromatographic separation resolves the target peptide from truncation products and from species carrying oxidised residues, while mass measurement confirms the expected molecular mass. Because the two common variants differ by roughly 280 daltons, a mass determination distinguishes them unambiguously. Purity is often quoted as a percentage of total peak area, although that figure depends on the detection wavelength and the integration method applied.
Reported half-lives differ widely between the two variants and between species. Values for the albumin-binding form are usually expressed in days, while the unconjugated form is measured in minutes to a few hours. Sampling schedules, assay sensitivity, and route of administration all influence the numbers, which limits direct comparison across studies. Whether sustained receptor occupancy produces different downstream effects from pulsatile stimulation remains an open question in the published work. Claims about relative potency should therefore be read alongside the specific study design that produced them.
Lyophilised powder is the usual supplied form. The material is hygroscopic, so vials are typically equilibrated to room temperature before opening in order to prevent condensation on the contents. Long-term storage is generally described at minus twenty degrees Celsius or colder, protected from light and moisture. Repeated freeze-thaw cycles are avoided because they promote aggregation and loss of soluble material. A reconstituted solution is considerably less stable than the dry powder and is normally kept refrigerated for short periods only.
Binding of the peptide to the growth hormone-releasing hormone receptor on pituitary somatotrophs triggers a G protein coupled cascade that raises cyclic AMP and opens calcium channels. The result is greater secretion of growth hormone into the bloodstream. Because the peptide acts at the same receptor as the natural hypothalamic hormone, its effect is amplified pulse size rather than an entirely separate release pathway. Receptor binding alone does not determine the response, since somatostatin tone and other inputs modulate the final output.
The albumin-binding version stays in circulation for days, because covalent attachment to serum albumin shields the peptide from rapid filtration and degradation. Reported half-lives for this form fall in the range of several days. The version without the linker is cleared in minutes, with estimates often near thirty minutes in animal work. These figures come from small studies and vary with assay method, species, and route, so they are best read as approximate rather than fixed constants.
Meglitinides help the pancreas produce insulin and are often called "short-acting secretagogues." They act on the same potassium channels as sulfonylureas, but at a different binding site. By closing the potassium channels of the pancreatic beta cells, they open the calcium channels, thereby enhancing insulin secretion. They are taken with or shortly before meals to boost the insulin response to each meal. If a meal is skipped, the medication is also skipped. Typical reductions in glycated hemoglobin (A1C) values are 0.5–1.0%.
=== Increased synthesis of ATF4 === In homeostasis, eIF2 combines with guanosine triphosphate (GTP) to activate the mRNA which will start transcription and simultaneously lead to the hydrolysis of GTP so that the process can start again. However during an essential amino acid shortage, P-eIF2α is phosphorylated and binds tightly to eIF2B preventing GDP from turning back to GTP leading to fewer mRNAs being activated and fewer proteins being synthesized. This response causes translation to be increased for some mRNAs, including ATF4, which regulates the transcription of other genes.
In 1979, the ANC's National Executive Committee had indeed endorsed a programme which included mass popular mobilisation inside South Africa, and its literature contained calls to reject the black local authorities. In addition, ANC president Oliver Tambo famously called for opponents of apartheid to make South Africa "ungovernable". On 8 January 1984, Tambo's annual statement to ANC members had included the entreaty:We should direct our collective might to rendering the enemy's instruments of authority unworkable. To march forward must mean that we advance against the regime's organs of state-power, creating conditions in which the country becomes increasingly ungovernable... Having rejected the community councils by boycotting the elections, we should not allow them to be imposed on us. We do not want them. We must ensure that they cease to exist. Where administration boards take over their functions, then these must be destroyed too.
macromolecule Any very large molecule composed of dozens, hundreds, or thousands of covalently bonded atoms, especially one with biological significance. Many important biomolecules, such as nucleic acids and proteins, are polymers consisting of a repeated series of smaller monomers; others such as lipids and carbohydrates may not be polymeric but are nevertheless large and complex molecules.

== Diagnosis == Lymphocytosis is usually detected when a complete blood count is obtained. If not provided the lymphocyte count can be calculated by multiplying the total white blood cell (WBC) count by the percentage of lymphocytes found in the differential count. The lymphocyte count can also be directly measured by flow cytometry.
== Patient interaction == Some radiologists, like teleradiologists, have no interaction with patients. Other radiologists, like interventional radiologists, primarily interact with patients and spend less time analyzing images. Diagnostic radiologists tend to spend the majority of their time analyzing images and a minority of their time interacting with patients. Compared to the healthcare provider who sends the patient to have images interpreted by a diagnostic radiologist, the radiologist usually does not know as much about the patient's clinical status or have as much influence on what action should be taken based on the images. Thus, the diagnostic radiologist reports image findings directly to that healthcare provider and often provides recommendations, who then takes the appropriate next steps for recommendations about medical management. Because radiologists undergo training regarding risks associated with different types of imaging tests and image-guided procedures, radiologists are the healthcare providers who generally educate patients about those risks to enable informed consent, not the healthcare provider requesting the test or procedure.
This occurred due to DOM being publicly distributed for free in the form of high-dose tablets by LSD distributor Owsley Stanley, who had personally learned of DOM from Shulgin. It is unclear why Shulgin provided information about DOM to Stanley, since doing so had the potential to risk Shulgin's professional career and the DOET clinical studies. One possibility is that Dow Chemical Company was not further looking into DOM and Shulgin thought that it was a promising drug that would otherwise be forgotten. In any case, street use of DOM was short-lived because the tablets caused a public health crisis due to them often producing very long durations (up to 3–4 days), intense experiences, worrying physical side effects, and hospitalizations. DOM was first reported on in the media and scientific literature in 1967 as a result of the crisis. DOM became illegal in the United States in 1968. Dow Chemical Company terminated its clinical research program on DOET due to the DOM public health crisis. DOET was subsequently first described in the literature by Snyder and colleagues in 1968. Snyder continued to be interested in DOET as a potential medicine, but it was never further developed. Snyder also described 2,5-dimethoxyamphetamine (2,5-DMA), which had been synthesized and tested by Shulgin, in the literature in 1968. DOM and DOET were further described in the scientific literature by Shulgin in 1969. In addition, Shulgin discussed DOM, DOET, TMA-2, and 2,5-DMA in a book chapter on hallucinogens published in 1970.
In June 2016 the Tritiated Water Task Force released a report on the status of tritium in tritiated water at Fukushima Daiichi nuclear plant, as part of considering options for final disposal of the stored contaminated cooling water. This identified that the March 2016 holding of tritium on-site was 760 TBq (equivalent to 2.1 g of tritium or 14 mL of pure tritiated water) in a total of 860,000 m3 of stored water. This report also identified the reducing concentration of tritium in the water extracted from the buildings etc. for storage, seeing a factor of ten decrease over the five years considered (2011–2016), 3.3 MBq/L to 0.3 MBq/L (after correction for the 5% annual decay of tritium). According to a report by an expert panel considering the best approach to dealing with this issue, "Tritium could be separated theoretically, but there is no practical separation technology on an industrial scale. Accordingly, a controlled environmental release is said to be the best way to treat low-tritium-concentration water." After a public information campaign sponsored by the Japanese government, the gradual release into the sea of the tritiated water began on 24 August 2023 and is the first of four releases through March 2024. The entire process will take "decades" to complete. China reacted with protest. The IAEA has endorsed the plan. The water released is diluted to reduce the tritium concentration to less than 1500 Bq/L, far below the limit recommended in drinking water by the WHO.
A/B testing is commonly employed when deploying a newer version of an API. For real-time user experience testing, an HTTP layer 7 reverse proxy is configured in such a way that n% of the HTTP traffic is routed to the newer version of the backend instance, while the remaining 100-n% of HTTP traffic hits the (stable) older version of the backend HTTP application service. This is usually achieved to limit the exposure of customers to a newer backend instance such that, if there is a bug with the newer version, only n% of the total user agents or clients are affected while others are routed to a stable backend, which is a common ingress control mechanism. == Development == The mod was developed by Adrian Finol in 2000. Finol created a unique mod that focused on intense, fast paced game-play that set it apart from other total conversion mods. Finol's goal was to create an online first person shooter that was more 'team based' than Counter-Strike, with bonuses for acting with team mates. Several versions were created under Finol's lead, each one adding new features and tweaking the game-play. In late 2001, Finol handed the FLF torch to Dave Dynerman so that he could start a new career with Valve Corporation. Several key contributors of the original FLF team now work for Valve. Under the lead of Dynerman, FLF went through several upgrades, resulting in even more versions. Dynerman went on to join Raven Software early in 2003 and Tony Sergi took over as the lead coder. Sergi created many versions for FLF, although not all became public. As the release of Half-Life 2 approached Sergi, along with the development team, faced a difficult decision. A great deal of time and work from all aspects had gone into the most recent unreleased versions of FLF (1.9 and Defiance), but the mod would have a hard time competing with other mods that were moving to the Half-Life 2 Source engine. Faced with a declining player base and only a handful of members left, the development team with real lifetime constraints for some, put further FLF work on hold.
As more African-Americans are incarcerated, the more the heuristic is enforced in the eyes of the courts, which contributes to yet higher imprisonment rates among African-American children. High numbers of African American arrests and charges of possession show that although most drug users in the United States are white, blacks are the largest group being targeted as the root of the problem. Furthermore, a study by Andrew Golub, Bruce Johnson, and Eloise Dunlap affirms the racial divide in drug arrests, notably marijuana arrests, where blacks with no prior arrests (0.9%) or one prior arrest (4.3%) were nearly twice as likely to be sentenced to jail as their white counterparts (0.4% and 2.3%, respectively). Harboring such emotions can lead to a lack of will to contact the police in case of an emergency by members of African-American communities, ultimately leaving many people unprotected. Disproportionate arrests in African-American communities for drug-related offenses has not only spread fear but also perpetuated a deep distrust for government and what some call racist drug enforcement policy. Additionally, a black-white disparity can be seen in probation revocation since black probationers were revoked at higher rates than white and Hispanic probationers in studies as published under the Urban Institute.
Generative artificial intelligence has been applied across multiple industries for content creation and automation. In healthcare, generative models are used for drug discovery and the generation of synthetic medical data to train diagnostic systems. In finance, they are used for report drafting, data generation, and customer service automation. Media and entertainment industries use generative systems for tasks such as music composition, script development, and image or video generation. Researchers and policymakers have raised concerns regarding accuracy, misuse, and impacts on academic and professional work.
About 60% of all casualties were civilians, who died as a result of disease, starvation, genocide (in particular, the Holocaust), and aerial bombing. The former Soviet Union and China suffered the most casualties. Estimates place deaths in the Soviet Union at around 23 million, while China suffered about 10 million. No country lost a greater portion of its population than Poland: approximately 5.6 million, or 16%, of its pre-war population of 34.8 million died. The Holocaust (which roughly means "burnt whole") was the deliberate and systematic murder of millions of Jews and other "unwanted" groups during World War II by the Nazi regime in Germany. Several differing views exist regarding whether it was intended to occur from the war's beginning or if the plans for it came about later. Regardless, persecution of Jews extended well before the war even started, such as during Kristallnacht (Night of Broken Glass). The Nazis used propaganda to great effect to stir up anti-Semitic feelings within ordinary Germans. After World War II, Europe was informally split into Western and Soviet spheres of influence. Western Europe later aligned as NATO, and Eastern Europe as the Warsaw Pact. There was a shift in power from Western Europe and the British Empire to the two new superpowers, the United States and the Soviet Union. These two rivals would later face off in the Cold War. In Asia, the defeat of Japan led to its democratization. China's civil war continued through and after the war, eventually resulting in the establishment of the People's Republic of China.
=== SAMDI-MS === While performing much of the early dynamic substrate and cell patterning work, Mrksich also pioneered an assay platform that utilizes SAMs of alkanethiolates on gold. The monolayers contain capture ligands (e.g. biotin or maleimide) that can selectively immobilize a peptide of interest. Subsequently, the monolayer can treated with a specific enzyme or a complex mixture, such as cell lysate, that can modify the peptide through various biological processes (e.g. phosphorylation). For quality control, the monolayers present these peptides against a background of tri(ethylene glycol) groups to prevent the nonspecific adsorption of protein to the surface that could obfuscate the reaction signal and, therefore, enable quantitative and reproducible assays. Most significantly, the monolayers can be characterized with MALDI mass spectrometry in a technique known as SAMDI-MS, which provides the masses of the substituted alkanethiolates and, therefore, the mass change of the immobilized peptide that results from enzyme activity. The method is compatible with standard array formats and liquid handling robotics, allowing a throughput in the tens of thousands of reactions per day. Importantly, the matrix-assisted laser desorption time-of-flight mass spectrometry (MALDI-TOF) analysis provides a fast and quantitative mass shift readout without the need for labels.

It is a synthetic peptide modeled on growth hormone-releasing hormone. The molecule is used in research on pituitary growth hormone secretion. It differs from the natural hormone through several stabilizing substitutions.
The DAC form carries a maleimide group that binds albumin and extends circulation time. The non-DAC form lacks this group and clears much faster. Both share the same receptor-binding core.
It targets the growth hormone-releasing hormone receptor on pituitary somatotroph cells. Activation raises cyclic AMP and promotes growth hormone release. Normal feedback pathways remain part of the response.
No. It is a synthetic analogue modeled on the first 29 residues of human GHRH. The substitutions and any linker are laboratory modifications rather than features of the endogenous peptide.
