Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
CJC-1295 is a synthetic analogue of growth hormone-releasing hormone, built on the 29-amino-acid GHRH(1-29) fragment. Four substitutions distinguish it from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes reduce enzymatic cleavage and extend the peptide's persistence in circulation. The compound is discussed in two forms, one carrying a drug affinity complex and one without it.
The drug affinity complex is a maleimidopropionic acid group attached to a lysine side chain. It reacts with the free thiol of cysteine-34 on circulating albumin, forming a covalent bond. This conjugation keeps the peptide in the bloodstream and shields it from rapid renal filtration and proteolysis. Reported circulation half-lives for the albumin-bound form fall in the range of roughly six to nine days in early human studies.
Reverse-phase high-performance liquid chromatography is the standard tool for purity assessment. The technique separates the target peptide from truncated or modified byproducts. Mass spectrometry confirms molecular weight and supports sequence verification. Electrospray ionization and matrix-assisted laser desorption are both used. Amino acid analysis provides an independent check on composition. Purity values are commonly reported as area percentage from the chromatogram. Residual trifluoroacetate and water content are also measured in many quality programs.
Batch-to-batch consistency depends on solid-phase peptide synthesis and subsequent purification. Coupling efficiency, resin choice, and cleavage conditions all affect the final profile. Counter-ion content and moisture can shift the apparent mass of a batch. Documentation typically includes a certificate of analysis with chromatograms and spectra. Independent verification by a second laboratory is sometimes requested. Whether a given certificate reflects the actual vial contents depends on chain of custody. Analytical methods themselves carry uncertainty that should be stated alongside results.
| Property | Value | Notes |
|---|---|---|
| Physical form | Lyophilized white powder | Supplied as a freeze-dried solid for research use |
| Molecular weight | Approximately 3,647 Da (DAC form) | Lower for the variant lacking the albumin-binding moiety |
| Solubility | Soluble in water and aqueous buffer | Typically dissolved in sterile or bacteriostatic water |
| Typical storage temperature | −20 °C or below | Dry powder, desiccated and protected from light |
| Common analytical method | Reversed-phase HPLC with mass spectrometry | Used for purity estimation and identity confirmation |
Reported half-lives differ widely between the two variants and between species. Values for the albumin-binding form are usually expressed in days, while the unconjugated form is measured in minutes to a few hours. Sampling schedules, assay sensitivity, and route of administration all influence the numbers, which limits direct comparison across studies. Whether sustained receptor occupancy produces different downstream effects from pulsatile stimulation remains an open question in the published work. Claims about relative potency should therefore be read alongside the specific study design that produced them.
Lyophilised powder is the usual supplied form. The material is hygroscopic, so vials are typically equilibrated to room temperature before opening in order to prevent condensation on the contents. Long-term storage is generally described at minus twenty degrees Celsius or colder, protected from light and moisture. Repeated freeze-thaw cycles are avoided because they promote aggregation and loss of soluble material. A reconstituted solution is considerably less stable than the dry powder and is normally kept refrigerated for short periods only.
Reconstitution is typically performed with sterile water or bacteriostatic water, added slowly against the vial wall. The resulting solution should be clear and colorless; cloudiness or visible particles suggest a problem with the material or the diluent. Once in solution, the peptide is less stable than the dry powder. Refrigerated storage at two to eight degrees Celsius is common for short-term holding, while freezing aliquots is described for longer periods.
Purity is most often assessed by reversed-phase high-performance liquid chromatography, reported as a percentage of total peak area. Identity is confirmed by mass spectrometry, which yields a molecular ion consistent with the expected sequence. Amino acid analysis and peptide mapping provide additional characterization. Reported purity values are method-dependent, so figures from different laboratories are not always directly comparable without details of column, gradient, and detection wavelength.
=== Electrolysis === Electrowinning and electrorefining respectively involve the recovery and purification of metals using electrodeposition of metals at the cathode, and either metal dissolution or a competing oxidation reaction at the anode.
The avian circulatory system is driven by a four-chambered, myogenic heart contained in a fibrous pericardial sac. This pericardial sac is filled with a serous fluid for lubrication. The heart itself is divided into a right and left half, each with an atrium and ventricle. The atrium and ventricles of each side are separated by atrioventricular valves which prevent back flow from one chamber to the next during contraction. Being myogenic, the heart's pace is maintained by pacemaker cells found in the sinoatrial node, located on the right atrium. The sinoatrial node uses calcium to cause depolarisation of the signal transduction pathway from the atrium through right and left atrioventricular bundle which communicates contraction to the ventricles. The avian heart also consists of muscular arches that are made up of thick bundles of muscular layers. Much like a mammalian heart, the avian heart is composed of endocardial, myocardial and epicardial layers. The atrium walls tend to be thinner than the ventricle walls, due to the intense ventricular contraction used to pump oxygenated blood throughout the body. Avian hearts are generally larger than mammalian hearts when compared to body mass. This adaptation allows more blood to be pumped to meet the high metabolic need associated with flight.
Detergents, such as SDS or Tween-20, which are used to break open cells and release the DNA. Protease enzymes, such as Proteinase K, which are used to digest proteins that may be binding to the DNA. Phenol and chloroform, which are used to separate the DNA from other cellular components. Ethanol or isopropanol, which are used to precipitate the DNA. Salt, such as NaCl, which is often used to help dissolve the DNA and maintain its stability. EDTA, which inactivates enzymes such as nucleases by chelating (binding) metals ions needed as cofactors. Tris-HCL, which is used to maintain the pH at the optimal condition for DNA extraction.
According to the study, however, when estimating the length of their partner's penis, most women would say a size significantly smaller than what their partner was recorded to be. This suggests that perception of size is not entirely accurate. The visual impression of the size is not necessarily in correlation with the feeling in the vulva and vagina. A very long penis can cause dyspareunia, if the man does not understand how to use it carefully.

The enzyme most responsible for estradiol 4-hydroxylation is CYP1B1. In humans, CYP1B1 mRNA and protein exhibit constitutive expression in the lung and kidney, as well as estrogen-regulated tissues such as breast, ovary and uterus. Whereas 4-hydroxylation constitutes the minor pathway in the liver, the greater proportion of CYP1B1 expression in extrahepatic tissues shifts the balance in favor of 4-OH-E2 formation. 4-OH-E2 is thought to be the most carcinogenic of all the estradiol metabolites, especially considering that CYP1B1 exhibits overexpression in breast cancer tumors. 4-OH-E2, like 2-OH-E2, can be physiologically active as well as tumorigenic. 4-OH-E2 is capable of binding ER with a reduced dissociation rate and prolonged activation, thereby inducing cellular growth and proliferation, adenohypophyseal hormone secretion, and prostaglandin production. Das et al. implicated 4-OH-E2 in the induction of estrogen-responsive genes, a response that exhibited partial or no abrogation by coadministration with an antiestrogen, providing evidence for the ability of 4-OH-E2 to carry out genetic upregulation via a pathway independent of ER signalling. Effects independent of ER binding include breakage of single-stranded DNA, especially when interacting synergistically with nitric oxide in human breast cancer cells and the production of quinones and free radicals. CYP1B1 can be induced by E2. ERα, after binding to estradiol, interacts with the CYP1B1 ERE to stimulate CYP1B1 expression.
=== Dependency === Adrenal suppression will begin to occur if prednisone is taken for longer than seven days. Eventually, this may cause the body to temporarily lose the ability to manufacture natural corticosteroids (especially cortisol), which results in dependence on prednisone. For this reason, prednisone should not be abruptly stopped if taken for more than seven days; instead, the dosage should be gradually reduced. This weaning process may be over a few days if the course of prednisone is short but may take weeks or months if the patient had been on long-term treatment. Abrupt withdrawal may lead to an Addisonian crisis. For those on chronic therapy, alternate-day dosing may preserve adrenal function and thereby reduce side effects. Glucocorticoids act to inhibit feedback of both the hypothalamus, decreasing corticotropin-releasing hormone (CRH), and corticotrophs in the anterior pituitary gland, decreasing the amount of adrenocorticotropic hormone (ACTH). For this reason, glucocorticoid analogue drugs such as prednisone down-regulate the natural synthesis of glucocorticoids. This mechanism leads to dependence in a short time and can be dangerous if medications are withdrawn too quickly. The body must have time to begin synthesis of CRH and ACTH and for the adrenal glands to begin functioning normally again. Prednisone may start to result in the suppression of the hypothalamic–pituitary–adrenal (HPA) axis if used at doses 7–10 mg or higher for several weeks. This is approximately equal to the amount of endogenous cortisol produced by the body every day.
The city's main opera house is Teatro Regio di Torino, where Puccini premiered his La bohème in 1896. It was burned down in 1936 and was rebuilt after World War II. On 8 October 2021, the European Broadcasting Union (EBU) and RAI announced that the city would host the Eurovision Song Contest 2022, following Italy's victory at the 2021 contest in Rotterdam, Netherlands, with the song "Zitti e buoni", performed by Måneskin. The contest took place at the Pala Alpitour, with both semi-finals of the contest took place on 10 and 12 May, and the grand final on 14 May. It was the first time that Turin has hosted the contest and the third time that Italy has hosted the contest overall, with the last being in Rome in 1991.
== Biosynthesis and industrial route == In terms of its biosynthesis, it is formed by the degradation of dihydrouracil and carnosine. β-Alanine ethyl ester is the ethyl ester which hydrolyses within the body to form β-alanine. It is produced industrially by the reaction of ammonia with β-propiolactone. Sources for β-alanine includes pyrimidine catabolism of cytosine and uracil.
=== Argentation chromatography === Ag+ binds alkenes reversibly, and silver nitrate has been used to separate mixtures of alkenes by selective absorption. This phenomenon is the basis of argentation chromatography. In some cases, alkene complexes can be isolated. The alkene can be liberated with ammonia.

== Synthesis == Pancreatic polypeptide is synthesised and secreted by PP cells (also known as gamma cells or F cells) of the pancreatic islets of the pancreas. These are found predominantly in the head of the pancreas.
== Agronomy == The plant thrives even in marginal soils and competes well with weeds. It is well-adapted to high-altitude subsistence agriculture and gives high yields; 30 tonnes per hectare are yielded at 3000 meters, but up to 70 tons per hectare have been produced under research conditions. Its extraordinary resistance to insects, nematode and bacterial pests is attributed to high levels of isothiocyanates. Although mashua is fully domesticated, it can persist in wild vegetation because of its aggressive growth and robustness. In Colombia, it is planted as a companion crop to repel pests in potato fields. Mashua’s high natural resistance to pests has made it a good crop for its potential in pest management. Glucosinolates contained in the plant have been shown to harm aphid herbivory. Spraying a crop with a mixture containing glucosinolates sourced from mashua can lead to up to 97% of aphid mortality. Molecules extracted from mashua can be part of a viable, effective, and eco-friendly alternative to synthetic pesticides. Traits like these raise the potential for mashua to be used in agroecology.
== Clinical significance == The loss of CD38 function is associated with impaired immune responses, metabolic disturbances, and behavioral modifications including social amnesia possibly related to autism. CD31 on endothelial cells binds to the CD38 receptor on natural killer cells for those cells to attach to the endothelium. CD38 on leukocytes attaching to CD31 on endothelial cells allows for leukocyte binding to blood vessel walls, and the passage of leukocytes through blood vessel walls. The cytokine interferon gamma and the Gram negative bacterial cell wall component lipopolysaccharide induce CD38 expression on macrophages. Interferon gamma strongly induces CD38 expression on monocytes. The cytokine tumor necrosis factor strongly induces CD38 on airway smooth muscle cells inducing cADPR-mediated Ca2+, thereby increasing dysfunctional contractility resulting in asthma. The CD38 protein is a marker of cell activation. It has been connected to HIV infection, leukemias, myelomas, solid tumors, type II diabetes mellitus and bone metabolism, as well as some genetically determined conditions. CD38 increases airway contractility hyperresponsiveness, is increased in the lungs of asthmatic patients, and amplifies the inflammatory response of airway smooth muscle of those patients.
== Mechanism == The total protein concentration is readout by an increase in absorbance at 565 nm, which can then be measured using colorimetric techniques, including using microplate readers. Most common reagents, except thiols and SDS, are compatible with the assay. An optimized formulation for the assay to maximize sensitivity in microplate format was described.

The two share the same 29-amino-acid backbone. CJC-1295 carries an additional albumin-binding moiety that markedly extends its residence time in circulation, while the form without that moiety clears faster. Literature sometimes applies the name loosely to either variant.
It has not been approved by major regulatory agencies for any therapeutic indication. Material sold under this name is typically distributed for laboratory research only. Clinical use is not supported by large controlled trials.
Early human studies of the albumin-binding form reported circulation half-lives on the order of several days. The value depends on assay method and study design. Estimates for the form without the binding moiety are considerably shorter.
The powder is normally held at minus twenty degrees Celsius or below. Light and moisture exposure should be minimized. Repeated warming and cooling cycles are avoided.
